Le but de ce projet est de comprendre les mécanismes qui contrôlent l'expression génétique histospécifique, et d'identifier les altérations qui conduisent à une expression aberrante dans les cellules tumorales. Le gène de l'alpha-foetoproteine (AFP) est utilisé comme modèle, en raison de son expression typiquement onco-foetale. Nous cherchons plus particulièrement à identifier les mécanismes assurant la non-expression du gène dans les cellules non-hépatiques, et ceux responsables de la cessation d'expression dans les cellules hépatiques adultes normales. [The aim of this project is to understand the basic controls of histospecific gene expression, and to identify the alterations leading to abnormal gene expression in tumour cells, using the alpha-foetoprotein (AFP) gene as a model. Expression of the AFP gene is typically onco-foetal. We focus on the identification of mechanisms controlling the lack of expression of the gene in non-hepatic cells, and the shut-off of expression in normal adult hepatocytes.]
